Summary:The purpose of this research was to know cross reaction Mecistocirrus digitatus protein with serum anti-Fasciola gigantica by using Western blot technique. The results can be used as a mecistocirrosis serologic diagnostic. This research was conducted using Mecistocirrus digitatus worms collected from the abomasums of large ruminant, especially cattle and Fasciola gigantica worms collected from liver of cattle. The worms were crushed and added with Phospate Buffer Saline (PBS) to make homogenates and then centrifuged at a speed 5000 rpm for 15 minutes. Homogenate Fasciola gigantica was immunized to mice to obtain serum anti-Fasciola gigantica at a dose of 200 μg / mice and booster 4 times with a span of two weeks. The homogenates was performed by SDS-PAGE technique to analyze the protein by using Commasie briliant blue staining. Cross reaction of Mecistocirrus digitatus protein with serum anti-Fasciola gigantica using by Western blot technique and obtained 6 protein bands are 124, 66, 49, 41, 34, and 27 kDa.
