Summary:Microorganisms are the most infection agent who cause diseases. The purpose of this study was to identify bioactive compounds, to test the activity and the potential for water-soluble extract of the yellow stem wood (Arcangelisia flava L.Merr) as antifungal against Candida albicans and Trichophyton mentagrophytes in vitro. The powder of kayu kuning stem refluxed for 2 hours with distilled water. The filtrate was evaporated and than counted the yield. Further qualitative and quantitative analysis by TLC using the comparative method berberine HCl. Antifungal activity assays performed with the agar diffusion method, and test the potential with microdilution method, then read absorbance at a wavelength of 595 nm. To facilitate the visual observation, it used MTT reagent, then the solution is still clear streaking in to agar media. Absorbance readings then analyzed by oneway ANOVA, followed by post hoc Tukey HSD test. The results provide yield of 5.70% w/w. At quantitative analysis showed the extract contains alkaloids berberine HCl by 1.55 ± 0.12% w/w. In the activity test, inhibition zone formed an average of C.albicans and T.mentagrophytes respectively 16.65±4,52 and 6.55±0,05 mm. The MIC values for both fungi are 10 mg/mL, the MBC values for C.albicans is 40 mg/mL and for T.mentagrophytes is 50 mg/mL . From the analysis with one-way ANOVA, shows that there are significant differences between the groups of test solution and the control group of fungi with p=0.020 for C.albicans and p=0.028 for T.mentagrophytes (p <0.050). The results of Post Hoc Tukey analysis, showed that both concentrations between groups and between groups of test solutions and positive controls did not differ significantly because the value of p> 0.050.
