الملخص:Background: Angiogenesis has a very important role on the development of liver cirrhosis. One of the specific stimulator of endothelial target cell in angiogenesis is VEGF (Vascular Endothelial Growth Factor). The common forms of VEGF gene polymorphisms are Single Nucleated Polymorphisms (SNP) or Insertion/Deletion (I/D). The research VEGF gene polymorphisms in patient with liver cirrhosis has not been established. Objective: to discover the difference in the proportion of I/D VEGF gene -2549 polymorphisms in patient with liver cirrhosis based on Child-Turcotte Pugh score. Method: This study uses cross sectional method with consecutive sampling. The total subject of the research is 38 patient with liver cirrhosis. 3cc of EDTA blood is collected as a sample from each patient. The sample would be examined in biochemistry laboratorium to know the proportion of I/D gene VEGF polymorphisms. The methods are DNA isolation, DNA amplification with Polymerase Chain Reaction (PCR), and electrophoresis. Kolmogorov-Smirnov test is conducted to investigate the significancy of difference in the proportion of I/D VEGF gene -2549 polymorphisms in patient with liver cirrhosis based on Child- Pugh Turcotte score. Result: The proportion of CPT A with DD genotype is 4 patients (10,5%), ID 5 patient (13%), and II 6 patients (15,9%). CPT B with genotype DD is 12 patients (31,6%), ID 6 patient (15,9%), and 3 patients (7,9%). CPT C with genotype DD is 6 patient (15,9%), II 1 patient (2,7%)and none CPT C patient with genotype ID. Significancy for CPT A with CPT B is 0,989, CPT A with CPT C is 0,356 and CPT B with CPT C is 0,785. Conclusion: There is no difference in the proportion of I/D VEGF gene -2549 polymorphisms in patient with liver cirrhosis based on Child-Turcotte Pugh score.
