總結:Promoters are one of the building blocks of a synthetically engineered biological system. Within a synthetically engineered biological system, they are involved in processes at the transcriptional level as they are responsible for the initiation of the transcription in gene expressions. The purpose of the study is to find effective and efficient ways to characterize the promoters so that it may be used in a predictable manner in future to aid researchers in designing novel synthetic genetic circuits. In this study we characterized the promoters based on two different models. The characterization was done in two different media to determine the relative promoter activity. And we did the experiment based on three constitutive promoters. Results have shown that both models will eventually provide us with the same result for relative promoter activity. And both models are able to provide us with accurate results of the relative promoter based on the comparison of the relative promoter activity results from Model 1 and Model 2 with the experimental results, which was then concluded to have no significant difference by the statistical Mann-Whitney U test. As such, both models are useful for the characterization of the synthetic promoters.
