Summary: | A series of experiments was conducted to evaluate the pharmacodynamic activities of ethanol-extracted Connarus grandis leaves (Connaraceae) at the Laboratory of Pharmacology FMIPA and the Laboratory of Pharmacology Toxicology IUC-Life Sciences Institute of Technology Bandung. The Hyppoeratie Screening method was employed in this study, then followed by spesific screening tests (e.g. toxicity, sleeping time, anti convulsion and hypotension) to increase the reliability of the Hyppoeratie Screening. Analyses of Hyppocratic Screening data by using a Lotus 1-2-3 program indicated, that the ethanol-extracted Connarus grandis leaves had sympatholitic and parasympathomimetic activities, and CNS deppressant and muscle relaxant effects. The intensity of effects increased linearly with doses (p<0.01). The ethanol-extracted Connarus grandis leaves was moderately toxic. The LD50 was 1.136 mg/kg bw for 6 hours, 0.467 mg/kg bw for 24 - 72 hours and 0.413 mg/kg bw for 6 days. There was a dose by time interaction on the weight gain of surviving mice (p<0.01). The ED50 of pentobarbital in mice was reduced by the 10 mg/kg bw extract from 29.06 mg/kg to 23.18 mg/kg bw. The extract prolonged the sleeping time of mice induced by pentobarbital doses of 25, 30 and 50 mg/kg bw (p<0.01). On the other hand, the onset of sleep was not significantly affected (p>0.10). The convulsion induced by 2 mg/kg bw strychnine nitrate was protected by extract with the CPD50 of 47.50 mg/kg and the DPD5O of 36.70 mg/kg bw, whereas the convulsion induced by 100 mg/kg pentetrazol was not.The doses of strychnine nitrate and pentetrazol above were CD100s. Thus, ethanol-extracted Connarus grandis leaves acted upon the inhibitory nervous system mediated by glycine.Determination of ethanol-extracted Connarus grandis leaves effect on rat left arterial carotid blood pressure using the Physiograph MK-III-S apparatus equipped with a pressure transducer indicated that, this blood pressure was significantly decreased (p<0.0005) for a long period (higher than 5 h). It was proven that ethanol-extracted Connarus grandis leaves was sympatholitic and/or parasympathomimetic. |
