Summary: | Folic acid is one of important molecules for infant growth, it can prevent NTD (neural tube defect), which can cause damage in brain and backbone of infant. Folic acid has low stability even under exposure in room temperature. To improve its stability is required special approach to protect its degradation during formulation and storage. The purpose of this study is to develop solid lipid nanoparticle to encapstulate folic acid. Solid lipid nanoparticle (SLN) of folic acid was prepared by microemulsion method. SLN consisted of palmitic acid as lipid, cremophor RH-40 as surfactan and ethanol as co-surfactan. To obtain good characteristic of microemulsion, we studied the influence of drug : lipid ratio and optimized concentration of surfactan and co-surfactan. SLN formed after the microemulsion was precipitated into water at temperature 5oC. Characterization of SLN included the SLN morphology , size and distribution size. The entrapment efficiency of SLN on folic acid indicating the loading capacity of SLN was also determined. The size and morfology of SLN was measured by Scanning Electron Microscope (SEM). Particle size distribution of SLN was measured by Particle Size Distribution Analyzer. Entrapment efficiency of SLN was measured by High performance liquid cromatography (HPLC). Transparent microemulsion and SLN were formed from 20% cremophor RH-40, 10% etanol, 0.015% folic acid and 2% palmitic acid. Particle size distribution of SLN suspension was in the range of 259-877 nm with entrapment efficiency of SLN was 17.28%. Various factors influenced SLN characteristic including type of lipid and surfactan, ratio of lipid, surfactan and drug in microemulsion, solidification process and solubility of drug in lipid. |
