Merr. bulbs as a cardioprotector has taken many interests to study its decreasing blood pressure effect through various mechanisms : diuretic effect, amplitude lowering effect, and inhibition in heart rate frequency. A 225 mg/kg BW of water extract and 112.5 mg/kg BW of ethanolwater extract were used in this research. In the diuretic test, the increase of urine volume was measured for 24 hours with furosemide 1.8 mg/kg BW as a standard group. In amplitude lowering test, NaCl 3.75 g/kg BW was used as an inductor and captopril 13.5 mg/kg BW was used as a standard. In the inhibition of heart rate frequency test, atenolol 9 mg/kg BW was used as a standard and adrenaline 1.2 µg/kg BW was used as an inductor. Volume of 24 th hour urine of furosemide, water extract, and ethanolwater extract group were significantly different (p<0.05) from control group with value 11.63 +- 4.04; 11.5 +- 1.98; and 13.2 +- 3.69 mL, respectively. At the 28 th days of the amplitude lowering test, all groups that has been treated with captopril, water extract, and ethanolwater extract showed decrease in amplitude with value (18.17 +- 6.09) x 10 2 ; (33.21 +- 16.56) x 10 2; and (30.14 +- 12.71) x 10 2 mV, respectively. Atenolol, water extract, and ethanolwater extract group showed no significant increase in heart rate frequency with 1.94 +- 0.27 Hz for atenolol group, 1.59 +- 0.27 Hz for water extract group, and 1.92 +- 0.33 Hz for ethanolwater extract group, after the intraperitoneal injection of adrenaline. It can be concluded that water and ethanolwater extract of Eleutherine americana (Aubl.) Merr. bulbs could increase the diuretic effect, decrease amplitude, and also inhibit the increase of heart rate frequency in Male Wistar rats. The change in these factors leads to blood pressure decrease. text
