The plants are known as sources of the valuable secondary metabolites, one of which is Morus cathayana. M. cathayana belongs to Moraceae family and its leaves are indispensable food for silkworm. This species was reported contained several types of secondary metabolites which exhibited important bioactivity such as Diels-Alder type adducts as antiinflamantory, 2-arylbenzofurans as antimicrobial, stilbenes and flavonoids as antihypertension. Phytochemical studies of M. cathayana revealed that many phenolic compounds have been isolated from the root bark and stem bark of the plant but none from the leaves. Furthermore, phytochemical study of the plant tissue culture of this species have not been reported. The aim of this research is to investigate phytochemical profiles from the leaves and the root culture of Morus cathayana. The leaves of this species were collected from Garut while the root culture was prepared by plant tissue culture technique and incubated in the liquid media, MS + IBA 1 ppm. From the leaves of M. cathayana yielded two arilpropanoid derivatives, MC-1 (propenilphenol derivative) and scopoletin (coumarin derivative). From liquid media of the root culture, afforded eight compounds, KMMC-1 (32 mg), KMMC-2 (32 mg), KMMC-3 (15 mg), KMMC-4 (4 mg), KMMC-5 (14 mg), KMMC-6 (15 mg), KMMC-8 (9 mg), and KMMC-9 (10 mg). The three of which, namely KMMC-1, KMMC-2, and KMMC-3, have been elucidated as new methoxylated Diels-Alder type adducts based on spectroscopic data including UV, IR and NMR. KMMC-1 showed a highly activity against leukemia murine cell P-388 (IC50 = 1.8 ug/mL), whereas KMMC-2 dan KMMC-3 showed low activity (IC50 = 8.8 ug/mL dan 5.8 ug/mL). text |
