Summary:Extraction and chromatographic methods are commonly applied in isolating alkaloid compounds from natural sources. In this research, a new isolation method of alkaloid was developed using caffeine and papaverine HCl as a model. Prior to alkaloid isolation, a selective reaction was performed to precipitate the alkaloids using salt solution of bismuth. The cation was then dissociated from the precipitate by adding sodium-ethylendiamintetraacetate. The method was optimized by finding molar ratio of the cation and finding an appropriate pH. It was found that each of 1 mole caffeine and 1 mole papaverine HCl required 6 moles and 9 moles of cationic bismuth respectively and an appropriate pH of the reaction is 2. The effectiveness of alkaloid isolation by this method was more than 90%. This method applied to isolate alkaloid from Nigella sativa L. seeds gave a yield of 0.13%.
