Summary: | Cellulose acetate ultrafiltration membranes with 30,000 Da Molecular Weight Cut Off (MWCO) has been prepared in this work. The composition of polymer casting solution is 17% wt cellulosa acetate, 24 wt % formamide and 59 wt% aceton with cold water as coagulants. The membrane has hidraulic permeability of 22.11 L/m2.h.atm and water flux of 52.8 L/m2.h at 2 atm. The cellulose acetate membrane has been used to concentrate crude a-amylase enzyme from aspergilus oryzae. The concentration process was caried out in a dead end ultrafiltration cell with the following condition: 2 atm applied pressure, 30-120 rpm stirring speed, temperature of 4oC, and addition 50 ppm Ca++ as stabilizer. This process gave the best result at stirring speed 30 rpm. Increase of enzyme activity and specific enzyme activity are 1.93 and 1.66 respectively with protein rejection of 66.80%, enzyme recovery of 96.74%, adsorbed protein of 367.4395 ug/cm2 and an enzyme flux of 8.60 L/m2.h. |
