Rat were grouped as: normal rat (normal group), gentamicin (100mg/kg/day) and piroxicam(3.6mg/kg/day) induced rat (positive control ), and gentamicin-piroxicam-induced rat followed by administration of corn silk extract (76.2mg/kg/day) (test group). At the 7th , 14th , and 21st day, five rats of each group were sacrificed to evaluate the alteration of renal. Gene expression of iNOS and MCP-1 were determined by RT-PCR and were quantified using imageJ relative to -actin expression. Results show that there were significant differences in renal histology between normal group and positive control, as well as between normal group and test group. While, there were no significant differences between positive control group and test group. RT-PCR showed that there were MCP-1 expression and revealed significant difference intensity at 14th day between positive control (1.23 + 0.119) and test group (0.65 + 0.283), p<0.05, but no significant difference at 21st day between control group and test group (0.59 + 0.333 and 0.65 + 0.210 respectively). On contrary, there were no iNOS expression was observed in all group. Accordingly, the administration of corn silk extract at dose 76.2mg/kg/day attenuated MCP-1 gene expression at 14th day significantly compared to untreated group. However, the administration of corn silk at that those could not improve renal histology. text |
