Summary:Free radical is one of the reason of degenerative diseases, it may be formed endogenously or environmentally. Free radical can be destroyed by antioxidants. Sechium edule leaves and seed have high potential antioxidant. The goal of the research is to isolate an antioxidant compound from labu siam leaves. Crude drug of labu siam leaves were consequtively extracted by maceration, using n-hexane, ethyl acetate and ethanol as solvents. Ethyl acetate extract was fractionated with vaccum liquid chromatography using silica gel 60 H and gradient eluent, containing n-hexane - ethyl acetate - ethanol. Purification was conducted by preparative thin layer chromatography. Characterization of isolated compound was performed by using ultraviolet-visible and infrared spectrophotometry. Isolation was done based on antioxidant activity test by using spectrophotometry and thin layer chromatography. Crude drug of labu siam leaves showed the presence of flavonoid, and steroid/triterpenoid compounds. Isolated compound had been isolated from the ethyl acetate extract. Isolated compound was supposed to be a flavonol with maximum absorbance at ? 268 and 362 nm, which has OH, aliphatic C-H, C=O (carbonil), and aromatic group. <br />
