The purpose of this research is to study the potency of N. tabacum and C. roseus cultures for quinine biotransformation. Callus initiation was performed on solid media with Schenk Hildebrant (SH) base media supplied with 2 mg/l NAA + 0,2 mg/l kinetin for N. tabacum and 1,5 mg/l 2,4-D + 0,15 mg/l kinetin for C. roseus. Cultures were mantained by subculturing every 3-4 weeks. Suspension culture was prepared by dispersing callus in liquid media. Biotransformation experiment was conducted by adding quinine 2 mg/ml for solid media and 1 mg/ml for liquid media. Biotransformation products were analysed by extracting cells and media, followed by thin layer chromatography (TLC) of the extracts. In both cultures, quinine was biotransformed but the process was very slow. New metabolites which supposed to be the result of quinine biotransformation were observed in both N. tabacum and C. roseus. Total new metabolite in C. roseus culture more various compared with N. tabacum culture. text
