Summary:Atherosclerosis is a major cause of artery coroner disease. One hypothesis said that atherosclerosis was caused by LDL oxidation which can develop plaque atheroma. Therefore, antioxidant activity of Eleutherine americana (Aubl.) Merr., known traditionally as cardioprotector, was performed using scavenging methods of DPPH free radical, in vitro and ex vivo lipid peroxidation, approaching prevention of atherosclerosis plaque formation. Ascorbic acid and a-tocopherol were used as standards. In scavenging DPPH free radical, the EC50 of water and ethanol-water extract of E. americana (Aubl.) Merr. were 462.44 ug/ml and 330.79 ug/ml respectively. In scavenging lipid peroxidation in vitro the EC50 of water and ethanol-water extract of E. americana (Aubl.) Merr. were 1774.01 ug/ml and 1426.35 ug/ml. In ex vivo lipid peroxidation scavenging test, dosage 225 mg/kg of water extract scavenged 25.18+3.5%, while 112.5 mg/kg dosage of ethanolwater extract scavenged 2.25+0.22%. Both water and water-ethanol extract of E. americana (Aubl.) Merr. showed weak antioxidant activity. Water extract of E. americana (Aubl.) Merr more potent in scavenging lipid peroxidation, while water-ethanol extract more potent in DPPH free radical scavenging.
