Summary: | Simethicone is a viscous liquid drug which is practically insoluble in water. It might cause difficulty to change simethicone into solid form or to tablet by using direct compression method. The purpose of this study was to change simethicone into solid form that could be tableted using direct compression method. In this study, simethicone was dissolved in methylene chloride (1:2 w/v). Hydrotalcite which is firstly calcined at 500oC for 6 hours and Avicel pH 102 were put into the solution with the composition 1:3, 1:5, and 1:7 (w/w). All compounds were mixed, then dried using oven at 30oC for 24 hours. Flow rate ability, density (bulk), density (tapped) and percent of compressibility of the compounds was characterized. Some of simethicone and hydrotalcite compounds were mixed with Avicel pH 102 and Mannitol ParteckTM. All compounds were tableted using direct compression method with the same position of upper and lower punch. The tablet then evaluated. Simethicone and calcined hydrotalcite compound with Avicel pH 102 as the diluent, produce better tablet with hardness 10,725 kg/cm2. This tablet showed the fastest disintegration time 10 minutes 17 seconds. Simethicone and calcined hydrotalcite compound have better compressibility than simethicone and Avicel pH 102 compound. Simethicone and calcined hydrotalcite compound showed the best compressibility at composition 1:7 with Avicel pH 102 as diluent. <br />
