The purpose of this research is to determine D-loop nucleotide sequences from four people of Sangiran. The method of this research consist of epithelium cell were used as DNA template, the DNA was then copied by PCR method, and the nucleotide sequences were determined by Sanger method. Agarose gel electrophoresis of PCR product showed that DNA sample size is 0.9 kb. The 451 nucleotides were determined and analyzed. The results were compared with Cambridge Reference Sequence (CRS) to find mutation positions in samples. At position 16226 were found the difference between nucleotide sequences of four samples and CRS. This difference is hypothesized as an identifying mark for nucleotide sequences of communities of Sangiran.This work is a part of database making for another research in finding relationship between people of Sangiran and Homo erectus fossil. text
