Summary: | Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis (M. tb). The difficult of medical treatment become resistance M. tb to antibiotics. Multidrug resistant tuberculosis (MDR-M. tb) is kind of resistant bacteria toward two kind of drug or more, usually in isoniazid (INH) and rifampin (RIF). INH resistance has been associated with G944C katG gene mutation. The previous research of isolate collection MDR-M. tb in Biochemistry Laboratory ITB was known that there are some isolate which have fenotipe characteristic resistant INH but is not G944C mutation, such as L3 MDR-M. tb isolate. This research objective to determine mutation of L3 katG gene fragment MDR-M. tb isolate which related with in fenotipe characteristic. The research include polymerase chain reaction (PCR), gel agarose electrophoresis, nucleotide sequence, and in silico analysis. PCR result is indicated one chain of DNA which size 0,43 kb and total of base in electroforegram is 416. Sequence aligment of L3 isolate MDR-M. tb and M. tb. H37Rv as standard showed some mutation in L3 katG gene fragment MDR-M. tb. This mutation cause modify of amino acid residue and estimated modify of KatG protein Structure. All of this mutation has not never been published. <br />
