Summary:The use of triphenyltetrazolium chloride as spray reagent for the chromatographic-spectrophotodensitometric analysis of methandrostenolone and dexamethasone has been studied. The adsorbant used was silica gel GF 254, and the developing solvent was a mixture of chloroform-metanol (9:1). The red color of triphenylformazan formed was stable for fifteen minutes for 0.10 ug dexamethasone and six hours for 1 ug dexamethasone. The detection limit for methandrostenolone was 7 ug and for dexamethasone was 0.05 ug. The red color of triphenylformazan of these two compounds had a maximum absorbance at 500 nm, and the recovery was 94.0 + 4.4 % for methandrostenolone and that for dexamethasone was 94.0 + 1.2 % . This method was then used for the determination of methandrostenolone and dexamethasone in plasma of healthy volunteers which were given a dose of 15 mg (three tablets of 5 mg) methandrostenolone and 2.5 mg (five tablets of 0.5 mg) dexamethasone.
