Microalgae cells obtained was then pretreated by the use of sonication or freeze drying, before their lipid content were extracted with organic solvents of n-hexane or chloroform:methanol (1:1). Total fatty acid content was determined by the method of Gravimetri, and the fatty acid characteristic was analyzed using the Gas Chromatography-Mass Spectrometry (GC-MS) method. The microalgae biomass concentration obtained from the culture using bubbling air or aeration with CO2 was 0.4903 and 0.3052 g/L, respectively. The total fatty acid content obtained from cells cultivated using bubbling air or aeration with CO2; cells pretreated by sonication or freeze drying; and cells extracted by solvents of n-hexane or chloroform:methanol (1:1) was 51.4 or 41.2%; 51.4 or 39.4%; and 37.3 and 39.4%, respectively. These result suggested that the maximum fatty acid production when microalgae cells was bubbling with air and then pretreated by sonication prior to extract their lipid using chloroform:methanol (1:1) as solvent. text
