SCREENING, ISOLATION, AND CHARACTERIZATION CELLULASE FROM MARINE BACTERIA

Cellulose is a sugar polimer built by glucose monomer linked by B-1,4-glycosidic bonds. This compound can be used as starting material for bioethanol production, which involves cellulose degradation into simple sugars. Cellulosa can be enzymatically hydrolyzed by cellulase. This enzyme may be produc...

This compound can be used as starting material for bioethanol production, which involves cellulose degradation into simple sugars. Cellulosa can be enzymatically hydrolyzed by cellulase. This enzyme may be produced by various microorganisms, such as fungi and bacteria. Cellulases from fungi are well characterized, but cellulases from bacteria remain unclear. However, duration time of cellulase production in fungi is longer than that in bacteria. Therefore, the screening of bacteria producing cellulase is important. This research reports screening of 33 marine bacteria using medium containing carboxymethylcellulose (CMC), optimation of cellulase production, and cellulase characterization. The screening of bacteria was performed in agar medium containing 1% (w/v) CMC as carbon source at 37oC. The CMCase activity was identified as clear zone surrounding bacterial colonies after the congo red dyeing. Two methods and variation of production medium were carried out to optimize the cellulase production. Batch and semicontinue fermentations were applied to get the best methods. Meanwhile, a variety of CMC and NaCl concentration was used to obtain the best media condition. <br /> text
