Description
Summary:Selection result toward two Gedongsongo hot springs bacteria showed that YTae-13 was more potensial bacteria for futher research. When compared to other isolate, YTae-13 had higher amount of colony when it was grown in agar medium and had wider clear zone diameter when it was cultivated in lipase's qualitative assay medium containing CaCl2 and tween 20. Previous 16S rRNA analysis result, showed that this bacteria has closest homology with Geobacillus lituanicus BGSCW9A89 which was confirmed to the results obtained from this research. Morphologically, this bacteria has a small size colony, concentric growth, convex surface, light yellow color, rod shaped-cell, and characterised as a gram positive bacteria. Growth curve character from 7,0 x 10(exponen 7) CFU/mL starter for a period of 21 hour showed two stationary phase at 11 and 17 hours after cultivation with growth rate of 0.153 h-1 and 0.0576 h-1 respectively. The comparison of growth-curve and enzyme activity curve, showed that three peaks of enzyme were observed at 9, 15 and 19 hour-old cells whose activity were 0.002566 unit/ug protein, 0.004164 unit/ug protein, and 0.005978 unit/ug protein. <br />
