Identification of Xylanase-Producing Bacteria Isolated from Worker Termite Reticulitermes santonensis

Xylanase is an enzyme which cleaves the B-1,4 backbone of xylan to produce xyloses and xylooligosaccharides. Xylanase has many applications, such as additive in livestock food; agricultural and industrial waste treatment; bioethanol production; chemicals production; and pulp and paper industries. In...

Xylanase has many applications, such as additive in livestock food; agricultural and industrial waste treatment; bioethanol production; chemicals production; and pulp and paper industries. In this research, the xylanase-producing-bacteria isolated from worker termite Reticulitermes santonensis were identified. The identification method was based on 16S rDNA sequence, using forward primer (Bactf1): 5'-AGAGTTTGATC(A/C)TGGCTCAG-3' and reverse primer (UniB1): 5'-GGTTAC(G/C)TTGTTACGACTT-3'. The result showed that 16S rDNA gene consisted of 1420 bp. In Silico analysis by using the BLAST programme and phylogenetic tree showed that the bacterium had 99% sequence similarity with that from Staphylococcus epidermidis strain MB (Gen Bank No. FJ768459.1), strain S09 (Gen Bank No. AY741152.1), and strain KL-096 (Gen Bank No. AY030342.1). The conventional identification of bacterium based on morphology-biochemistry characteristics showed that the bacterium belonged to Staphylococcus arlettae. Based on these results, the bacterium isolated from worker termite R. santonensis was Staphylococcus sp. <br /> text
