Nicotine patch (NP) is one type of therapies to stop smoking. In this study, NP was formulated and evaluated. Nicotine gel formula consisted of HPMC as a gelling agent, sodium benzoate as a preservative, and polyisobutylene (Oppanol B100) as an adhesive. In vitro permeability of nicotine from NP was evaluated using flow-through diffusion cells for 24 h and membrane used was shed Python reticulatus skin. Stability of nicotine in NP was evaluated at room temperature for 90 days. Formulation of NP consisted of HPMC 10% in water, sodium benzoate 0.25%, and polyisobutylene 1 g (2.5% in n-hexane) as an adhesive. Nicotine permeated from NP was 21.89 +- 0.23%, these results were comparable to the commercial preparation (23.22% +- 0.24%). Meanwhile, the use of phosphate buffer pH 6.5 as a dispersion medium of HPMC increased the cumulative amount of nicotine permeated from NP (50.13 +- 0.92%). No irritation signs were observed (n=3). Nicotine in NP was stable (98.87 +- 2.66%) for 90 days of storage. NP formulation in this study resulted comparable nicotine permeation profile with commercial product, chemically stable and no sign of irritation. Modification of HPMC dispersion medium from aquadest to phosphate buffer increased permeated amount of nicotine from NP. text
