Summary: | Chemistry content (secondary metabolites) of Andrographis paniculata has a lot of pharmacological activities. Regulation of secondary metabolites production of this plant is expected to increase the use of it for medicinal purposes. Combination of tissue culture and elicitation is the techniques that could be used for optimizing the secondary metabolite gain. This research aimed to study the effect of elicitor derived from Saccharomyces cerevisiae, as a common elicitor, on secondary metabolite of A. paniculata callus cultures. Callus was initiated from leaf and grew optimally on Murashige and Skoog (MS) medium with the addition of 0.5 uM 2,4-dichlorofenoxy acetic acid (2,4-D) and 0.1 uM benzyl amino purine (BAP). Callus in the 12th week of age was elicited with yeast extract that dispersed in MS medium at concentrations of 0.05%, 0.25%, and 0.50%. The harvesting time were 0, 2, and 6 days after elicitation. The dried callus and crude drugs of wild type was extracted with methanol. These extracts was analized qualitatively using thin layer chromatography (TLC). The results showed the different TLC profile between methanol extract of callus and original plant, that indicated the production of different secondary metabolites. It can be concluded that the elicitated of yeast, in the medium of growth, to the callus influenced secondary metabolite synthesis in cultured callus and elicitated callus and the real plant. |
