Microalgae are a photosynthetic organism producing biopigments continuously and abudantly. The aim of this research is to isolate and characterize biopigments from marine microalgae Nitzschia closterium. Methods of the research involved cultivation of microalgae, extraction of biopigments, separation of biopigments, and characterization of biopigments. Wet biomass obtained from 1 L culture in Walne Medium for 4 d was 0.92 g. Crude extract pigment (25 mL) was obtained from extraction of 4 g dried biomass in 32 mL acetone. Separation of pigments by thin layer chromatography (TLC) with eluent acetone:hexane (3:7) resulted in 10 different pigments containing trans-β-carotene (maximum absorbtion at 452 and 481 nm), chlorophyll a (409 and 662 nm), lutein (429 and 456 nm), and fucoxanthin (446 nm). Two biopigment fragments were identified as phaeophytin a and phaeophytin b with retention factor value (Rf) of 0.6 and 0.4 respectively. The remaining biopigments could not be identified. The results indicate that marine microalgae Nitzschia closterium can be use for a source of photosynthetic pigments. text
