Sunflower (Helianthus annuus) is one of the species belonging to Asteraceae family which is also commonly used as medicines. This species is often applied to treat malaria disease in many countries, such as in Indonesia, India, and China. However, the antimalarial bioactive components contained in H. annuus leaves have not yet reported. In addition, the secondary metabolites in this plant have also been known to have anticancer activities. But there are no recent studies have been reported about the anticancer activity against murine leukemia P-388 cells. Therefore, the objective in this research was to isolate the secondary metabolites from H. annuus leaves using extraction methods and various chromatographic techniques, such as vacuum liquid chromatography, radial chromatography, gravity column chromatography, and Sephadex LH-20 column chromatography. Furthermore, the extract and secondary metabolites isolated were examined on antimalarial and cytotoxic against murine leukemia P-388 cells assays. Nine compounds have been successfully isolated from methanol extract of H. annuus leaves. Among them seven compounds were elucidated according spectroscopic data including, UV, IR, NMR (1H, 13C, HSQC, HMBC, and COSY), and MS spectral data. The compounds were identified as demethoxyencecalin, nevadensin, xanthomicrol, demethoxysudachitin, the mixture of acerosin and sudachitin, and 16&#946;,17-dihydroxy-ent-kaurane-19-oic acid. Moreover, the two other compounds based on UV spectral data were suggested as a benzopyran derivatives. The preliminary assay of methanol extract of H. annuus leaves on antimalarial and cytotoxic activity against murine leukemia P-388 cells showed IC50 values were 6.0 &#956;M and 17.5 &#956;g/mL, respectively. One of the compounds, i.e. demethoxysudachitin has been examined on antimalarial assay and showed high activity with IC50 values was 1.85 &#956;M.
