This is caused by high levels of biodiversity, one of them as a source of plants that could be developed into a wide range of herbal medicines that have significant economic value. Zingiber cassumunar (Zingiberaceae) is one of the plants that have been reported to have efficacy as reducing obesity, fever, sedative, laxative, and vermifuge. These plants are distributed in the tropics to the subtropics, with a center distribute in Southeast Asia. In Indonesia, this plant is locally known as "Bangle". Based on research in India, Japan, and Thailand to the species Z. cassumunar, reported that the main content are essential oils (monoterpene and sesquiterpene), diarylheptanoids (curcuminoids), phenylbutanoids, saponins, and flavonoids. In addition, Z. cassumunar been reported to have important bioactivity such as anti-inflammatory, antioxidant, antimicrobial, antiallergy, and cytotoxic. However, research on the secondary metabolites of Z. cassumunar from Indonesia is still definite. Based on the many benefits and diversity of secondary metabolites is dependent on the place where the plant grows, the study aims to isolate the secondary metabolites of Z. cassumunar and to characterize. In this study, separation and purification methods performed by vacuum liquid chromatography and radial chromatography; characterization of the secondary metabolites using mass spectroscopy and NMR spectroscopy [1D-NMR(1H-NMR and 13C-NMR) and 2D-NMR (HSQC, HMBC, COSY, and NOESY) ]. As a <br /> <br /> <br /> <br /> <br /> <br /> <br /> result, five compounds have been isolated from the methanol extract of Z. cassumunar, namely: the three compounds of non-polar fraction [pure compound (E)-4-(3',4'- <br /> <br /> <br /> <br /> <br /> <br /> <br /> dimethoxyphenyl)but-3-en-1-yl acetate; a mixture of two compounds (E)-4-(3',4'-dimethoxyphenyl)but-3-en-1-yl oleate and (E)-4-(3',4'-dimethoxyphenyl)but-3-en-1-yl <br /> <br /> <br /> <br /> <br /> <br /> <br /> linoleate] and two pure compounds of the polar fraction [(E)-4-(3',4'-dimethoxyphenyl)but-3-en-1-ol and 1,4-epoxy-p-mentan-2-ol]. Two phenylbutanoids [(E)-4-(3',4'- <br /> <br /> <br /> <br /> <br /> <br /> <br /> dimethoxyphenyl)but-3-en-1-yl acetate and (E)-4-(3',4'-dimethoxyphenyl)but-3-en-1-ol] has previously been reported to be isolated from the same species; two fatty acid ester <br /> <br /> <br /> <br /> <br /> <br /> <br /> phenylbutanoids [(E)-4-(3',4'-dimethoxyphenyl)but-3-en-1-yl oleate and (E)-4-(3',4'- dimethoxyphenyl)but-3-en-1-yl linoleate] have been reported previously but was isolated <br /> <br /> <br /> <br /> <br /> <br /> <br /> from the artifacts prasaplai (Thai traditional medicine, which consists of a mixture of rhizomes Z. cassumunar and Nigella sativa seeds); 1,4-epoxy-p-menthan-2-ol for the first time isolated from the rhizome of Z. cassumunar, but has previously been reported to be isolated from the rhizome of other species, namely: Ferula jaeschkeana (Apiaceae) and Jatropha isabelli (Euphorbiaceae).
