Summary:Indonesia is one of three countries that have many plants which has 30.000 species. The presences of secondary metabolites such as alklaoid, flavonoid, quinon, lignin, steroid, and terpene make it as source of medicine. Eupatorium inulifolium or “Kirinyuh” had reported that it contains terpene and flavonoid. This plant also have many function such as antimalarial, pesticide, and wound drug. Base on those facts, the aims of this research are the <br /> <br /> <br /> <br /> <br /> <br /> <br /> isolation and characterization of the secondary metabolits of it leaves. This study covers the process of maceration 1.94 kilograms of dry leaves powder for 2 x 24 hours using the MeOH. Twenty grams of MeOH extracts were separated by various separation techniques such liquid vacuum chromatography, radial chromatography, and sephadex column <br /> <br /> <br /> <br /> <br /> <br /> <br /> chromatography to produced pure compounds. The molecular structure of pure compounds was determined by the data of UV-Vis spectrum and NMR (1H NMR, 13C NMR, HSQC, HMBC, and COSY). Two compounds derived from flavonoids and terpenoids have been isolated and identified, which are 5,7,3’,4’-tetrahydroxy-6-methoxyiflavone (= nepetin) from flavonoid and inulifolinone E from norlabdane diterpene. A feature of 5,7,3’,4’-tetrahydroxy-6-methoxyiflavone are presence of methoxy groups at C-6 from ring A of flavone compound. This case is rarely found in other plants, but from genus Eupatorium such as E. inulifolium, E. arnottianum, and E. littorale. Inulifolinone E is characterized by the presences of furan ring and conjugated double bond of the carbonyl group at position C-12, exocyclic alkenes at position C-8, two hydroxyl groups, and one monoasetat ester. Based on these results, in this study have been isolated and identified two compounds, namely 5,7,3',4'-tetrahydroksy-6-methoxyflavone (=nepetin) and inulifolinone E. Both of these compounds have previously been found and reported.
