Summary: | Biosurfactant is surfactant produced by living microorganisms as secondary metabolite. The purpose of the present work is to produce biosurfactant from halophilic bacteria Pseudomonas stutzeri that has been isolated from a mud cratter Bledug Kuwu, Purwodadi, Central Java. The potential of P. stutzeri in producing biosurfactant was examined with haemolytic assay by using blood agar medium. The clear zone was observed indicating that cell blood wer lysized by biosurfactants produced by bacteria. Biosurfactant activity reached the highest level when the culture medium for production contained olive oil and KNO3 as carbon and nitrogen sources, respectively, and shaking incubated at 37 oC with speed 150 rpm for 20 hours. At this condition , the resulted biosurfactant was able to reach 3.5 cm in the diameter of oil spreading test and 100 % of emulsification index. Various physicochemical test showed that the biosurfactant reach highest activity at 70 oC, pH 12, and when solution contains 17 % (w/v) NaCl. The resulting biosurfactants also passed CTAB-methilen blue test suggesting that it is most likely rhamnolipid type of biosurfactant. Structural elucidation by infrared spectrophotometer was able to identified several functional group-vibrations:O-H at 3455.52 cm-1, H-C=O at 2847.79 cm-1, C = O at 1761.2 cm-1, C-O-C esther at 1473.06 cm-1, dan C-O ether at 1169.29 cm-1. Those functional groups mathced with most rhamnolipid structure. |
