Summary: | Salmonella typhi cause human typhoid fever. CarA gen of S. typhi is one of genes predicted to be expressed more in vivo and important to its pathogenesis. Expression level of CarA gen of S. typhi in this research studied by immunology approach. Terminal carboxy and terminal amino rabbit antipeptide antibodies of CarA protein of S. typhi were succesfully produced each from synthetic peptide (NH3)—Lys— His—Tyr—Arg—Ser—Ser—Ala—(COOH) and (NH3)—Ser—Ala—Leu— Leu—Val—Leu—Glu—Lys—(COOH). The purpose of the present study would further characterize the antipeptide antibodies from cross-reaction aspect. Antipeptide antibodies of CarA protein of S. typhi cross-reaction were examined by homology study in si;ripto and experimentally using ELISA and Western Immunoblotting technique. Amino acid sequence of another bacteria which have homology and predicted to exhibit crossreaction with antipeptide antibodies are found. Among the bacteria, there are three bacteria which have spesific homology is selected : Salmonella typhimurium, Escherichia coil, and Pseudomonas aeruginosa. ELISA and Western Immnnoblouing technique showed cross-reaction of terminal carboxy and terminal amino antipeptide antibodies of CarA protein of S. typhi with S. typhimurium, E. coil and P. aeruginosa. Cross-reaction level of terminal carboxy antipeptide antibodies with S typhimurium, E coil and P. aeruginosa are 74,7%, 34,4% and 186,8%, while terminal amino antipeptide antibodies are 104,7%, 71,2%, and 58, 1% respectively. It is expected that terminal carboxy and terminal aminoantipeptide antibodies of CarA protein of S. typhi could be used for study of CarA protein of other organism based on cross-reaction level differentiation of terminal carboxy and terminal amino antipeptide antibodies of CarA protein of S. typhi. |
