The present research aims to construct chromatography column based on matrix of frustule from N. closterium for protein separation. The methods included isolation of the frustule by biomass cleaning with HCl and baking at 700 ºC, identification of the frustule morphology and chemical composition by scanning electron microscope energy dispersive spectroscopy (SEM-EDS), and separation of protein mixture (BSA and lysozyme) by frustule matrix column. The results showed that yield of frustule obtained were 7.25±0.61% (g/g wet biomass). The frustules of N. closterium had a penates or needle-like in shape with pore size in a range of 20–30 nm. The SiO2 content of the frustule was 73.66% (w/w). The separation of mixtures of lysozyme and BSA by chromatography column based on frustule matrix produced two fractions (No 19 and 30). SDS-PAGE analysis of fractions showed that a protein band of 66 kDa indicating BSA was observed from fraction No. 19, meanwhile no protein band observed from fraction No. 30. This showed that frustule of N. closterium could be applied as a matrix of chromatography column. text |
