The objective of the research is to utilize the biosilica cell wall of Navicula sp. as a matrix for protein purification. The methods included production of Navicula sp. cells in enriched seawater medium, isolation and purification of biosilica by acid treatment as well as baking of biomass, characterization of biosilica morphology by SEM-EDS, and application of biosilica matrix to purify mixtures bovine serum albumin (BSA) and lysozyme. The results showed that the growth of Navicula sp. cells with an initial concentration of 200,000 cell/mL was to be a maximum density of 4.47 g/L for 8 d. The amount of biosilica mass obtained from wet biomass was 1%. The frustule of Navicula sp. was boat-like oval possessing porous sizes between 100–200 nm, with silica (SiO2) content of 94.7%. Characteristics of biosilica frustule of Navicula sp. was suitable used for a matrix of filtration gel column. Separation of mixtures BSA and lysozyme using a biosilica column produced protein fractions. SDS-PAGE analysis showed a protein band of ~66 kDa of fractions 15 and 16 identified as BSA fraction, whereas lysozyme was not observed. text
