One of the ways to remove organohalide pollutants is by bioremediation. Bioremediation is the way of reducing pollutants by using microorganisms. Pseudomonas is one of Gram negative bacteria that is known for its ability to use organohalide compounds as the sole carbon source. This research aims to isolate and characterize monochloroacetic acid (MCA) degrading Pseudomonas from organohalide contaminated soil. Isolation was carried out using some specific media for Pseudomonas with MCA as the sole of carbon source. Some of the isolates obtained were characterized by their growing ability at various MCA concentrations. The bacterial characterization was performed using a 16s rDNA typing technique through PCR using Bact27F as a forward primer (5'-AGA GTT TGA TGG CTC AG-3 ') and UNI1492R as a reverse primer (5'-GGT TAC CTT GTT ACG ACT T-3'). The ability of isolates to degrade MCA were determined by measuring the amount of Cl- released to the medium using Bergmann and Sanik methods. This research yielded 8 Gram negative isolates, one of which was able to grow in a minimum medium containing 20 mM MCA and able to degrade 4,85% MCA. Ribotyping has been successfully identifying four isolates which were Pseudomonas aeruginosa, Pseudomonas sp., Alkaligenes sp., and Serratia rubidaea. text
