Bark has a high lignin content, around 43%. Thus, it cause saponin extraction from the bark become harder. In this experiment, we do a pretreatment delignification to decrease the lignin content using Phanerochaete chrysosporium with incubation day variation 7, 14 and 21 days and see the effect to saponin yield from the bark. Analysis for lignin content was done by Chesson-Datta method, and analysis for total saponin was done using extraction. Incubation for 21 days could degrade 15% lignin with the final content in bark was 36,65 + 1,66 %. With this, we could increase saponin yield from extraction about 7,5 fold from normal, it was 7,02 + 0,62 % g/g dry weight. This result shows that the longer incubation could make the extraction easier and resulting the saponin yield higher. text
