Naturally, the wild plant produces a small amount of this secondary metabolite. Therefore, in vitro culture of adventitious roots offers an alternative way to increase the production of saponin. This study was performed to investigate the effect of methyl jasmonate (MeJA) and salicylic acid (SA) on the adventitious root growth and saponin content. This experiment was designed to test 4 different concentrations (0, 0.05, 0.1, and 0.2 mM) using two different elicitors, MeJA and SA and incubated for 5, 10 and 15 days. Initially, the root culture was induced on solid Murashige and Skoog (MS) medium supplemented by IBA and NAA to determine the optimum growth for adventitious roots. The result showed that IBA 10 μM produces the highest biomass among other treatments (0.035 g/day of growth rate). Using this optimum condition, adventitious roots were incubated in ½MS liquid medium in flask culture added by IBA 10 μM and different concentrations of MeJA/SA. Despite inhibition on root growth, saponin production increased upon elicitation with MeJA and SA with concentration 0.2 mM for 15 days about 1.5 and 1.3 times compared to control. Therefore, elicitation with MeJA was better than SA for saponin production in javanes ginseng. text
