The root culture was elicited with elicitor derived from autoclaved P. aphanidermatum at concentrations of 0.05, 0.5, 1.0 and 5.0 mg DW/ mL then harvested at 18, 24, 36, 48, and 72 hours after elicitation. The ajmalicine was analyzed qualitatively and quantitatively by using HPLC (High Pressure Liquid Chromatography) connected to chromatopack CR-7A plus. The result of elicitation showed that ajmalicine content was influenced significantly by concentration and harvesting time. The highest ajmalicine content on the roots (165,609 + 1,141 DW/g ) with the percentage of increment 181.633% was elicited by addition of 5.0 mg DW/mL elicitor, for 24 hours, while the highest ajmalicine content on the media (291,473 ± 0,872 mg DW/ L) with the percentage of increment (306, 466%) was elicited by addition of 5.0 mg DW/mL elicitor for 36 hours. This result indicated that elicitation might be responsible for further release of ajmalicine content into the media rather than its accumulation in the root. text
