Summary: | Salmonella typhimurium is bacterial pathogen which has served as a model for investigation on typhoid disease. Mekalanos, etal have discovered genes of S. typhimurium which that expressed in vivo maximally and expressed in vitro minimally . This research aim to examine the expression level of S. typhimurium CarA protein in vivo and in vitro. The research strategy is through immunology approach by using ELISA and Western immunoblotting methods. The in vitro result by using ELISA obtained A4os = 0,217 for 300 ng antygen and found band on 45 kDa using Western immunoblotting. The result show that antybodies were able to identify S. typhimurium CarA protein. By ELISA mice CarA protein present low absorbant and so supported by Western immunoblotting which don't give band, it means that there is no cross reaction with the mice CarA protein. Detection S. typhimurium CarA protein in vivo expression by ELISA technique in 1,7 mg mice spleen infection contain about 2.1 x 108 cells give absorbent higher than in vitro expression for the same total cells. In 2,7 mg pain mice tissue investigated by Western immunoblotting give a weak band. This fact indicates that the expression level of S. typhimurium CarA protein in vivo is relatively higher than in vitro by immunology approach that need to be more completed. So it need another strategy to support that fact. |
