Skim latex with dry rubber content <br /> <br /> <br /> <br /> 4,02% (w/w) used in this study was obtained from natural rubber Hevea brasiliensis <br /> <br /> <br /> <br /> latex by using combination of 10% (v/v) sodium alginate creaming and low speed <br /> <br /> <br /> <br /> centrifugation method. A. oryzae culture was cultivated for 5 days (30oC, 150 rpm) <br /> <br /> <br /> <br /> in Potato Dextrose Broth 26,5 g/L medium and skim latex serum medium with <br /> <br /> <br /> <br /> different concentration 25, 50, 75, 100% (v/v) to evaluate the effect of skim latex <br /> <br /> <br /> <br /> serum concentration to the growth and emulsification activity. The best result was <br /> <br /> <br /> <br /> obtained through Potato Dextrose Broth medium combined with 100% (v/v) skim <br /> <br /> <br /> <br /> latex serum, yielding biomass 7,21 g/L and emulsification index (E24) 48,28%. <br /> <br /> <br /> <br /> Furthermore, growth and biosurfactant production profile were plotted through 6 <br /> <br /> <br /> <br /> days fermentation (26oC, 150 rpm). Optimum biosurfactant concentration up to <br /> <br /> <br /> <br /> 1,00 g/L (E24=45%) was obtained from Potato Dextrose Broth medium combined <br /> <br /> <br /> <br /> with 100% (v/v) skim latex serum at the sixth day, 72% higher compared to skim <br /> <br /> <br /> <br /> latex serum medium without Potato Dextrose Broth, showing significant effect of <br /> <br /> <br /> <br /> glucose supplementation to biosufartant production. Biosurfactants produced were <br /> <br /> <br /> <br /> characterized using fourier transform infrared spectroscopy and exposed the <br /> <br /> <br /> <br /> presence of carbohydrate, lipid and protein groups or called glycolipopeptide. <br /> <br /> <br /> <br /> Biochemical analysis shows the ratio of 1,8:17,2:0,6 for skim latex serum medium <br /> <br /> <br /> <br /> and 3,6:13,9:1 for Potato Dextrose Broth combined with skim latex serum medium <br /> <br /> <br /> <br /> respectively <br /> <br /> <br /> <br /> <br /> <br />
