Summary: | Stevia plant leaves (Stevia rebaudiana Bert.) contain sweetener compounds in the form of rebaudiosida-A which has high economic value. Conventional propagation is inefficient to meet the needs on industrial scale. Therefore, superior stevia plantlet propagation is produced with alternative in vitro culture method. Moreover, the enhancement of the rebaudioside-A compound can be performed with drought stress, which can be mimicked by shorter immersion time of plantlet in the medium. In this study, cultivation of stevia shoot was performed on TIS RITA® bioreactor in order to evaluate the effect of different immersion time on biomass and rebaudioside-A content. The stevia shoots were initiated on MS medium and cultured on the RITA TIS bioreactor with ½ MS medium. The parameters measured were growth, consumption rate of sucrose and ions, the content of rebaudioside-A with HPLC. The results showed that the duration of immersion for 30 minutes every 6 hours resulted in higher dry biomass weight than the duration of immersion for 15 minutes every 3 hours. The highest rebaudioside-A content was obtained at immersion for 30 minutes every 6 hours compared to immersion for 15 minutes every 3 hours, 1.89 mg / g and 1.17 mg / g respectively. |
