Summary:<p align="justify">Brightfield illumination microscopy is the basis of almost all current optical microscopes and used by many people on many different fields. In this technique the object will appear darker than the bright background. The contrast that resulted from this technique relies on the interaction between light and objects, such as differences in absorption, reflection, diffraction, polarization and refraction of light by the object. However, this technique produces poor image contrast in objects that have a transparent structure, and so we need another method to increase the contrast of the image of the object. In this experiment three methods was carried out to enhance the image contrast, those are darkfield microscopy, histogram equalization and combination between darkfield microscopy with histogram equalization method. In darkfield microscopy, the middle of light source will be blocked using patch stop that is placed under the condenser lens. It will cause only oblique rays that pass through the object, the result is the image that is formed makes the object look bright with a dark background. At this research patch stop is created manually using a solid but thin material such as cardboard, duck tape, and rubber. Histogram equalization (HE) method uses the image histogram to improve the image contrast so the image becomes better by reconstructing its pixel intensity In this research, contrast limited adaptive histogram equalization method (CLAHE) is used. This method is a developed version of standard histogram equalization method. The image is processed with HE taken from a microscope with brightfield microscopy methods, furthermore the image will be processed using MATLAB software. Each image contrast value is calculated and processed using ImageJ software. In this research, the contrast and imaging result from the three methods are analyzed. Afterwards, the analysis result of those methods are compared to determine which method that shows the best outcome.<p align="justify">
