Summary: | Two proteins were believed to be involved on the termination translation process, namely eRF1 and eRF3, in eukaryotic system. The proteins were coded by SUP45 and SUP35 gene respectively. Naturally, the expression of both proteins in Saccharomyces cerevisiae are very low. To obtain high quantity of the proteins, heterologous expression of proteins in Ercherichia coli was carried out through pUKC630 and pUKC1803 plasmids. The proteins were purified through IMAC system. The purified eRF1 with the above method was used on purification of anti eRF1 antibody. In other hand, partial purified eRF3 was further purified by electroelution before used as antigen to induce anti eRF3 antibody. Characterization of anti eRF3 and anti eRF1 antibodies was carried out by Western blot analysis. The result showed that specificity of anti eRF3 and anti eRF1 antibodies were relatively high. |
