Summary:Patchouli (Pogostemon cablin, Benth) is essential oil-producing plants that have high economic value. Quality of patchouli oil can improved by using superior seeds which can produced by in vitro propagation method. In addition, drought stress can increase levels of patchouli alcohol in patchouli oil. In this experiment, patchouli biomass propagation is conducted by using TIS RITA bioreactor. Production biomass, yield of essential oil, and essential oil content, were evaluated by cultivating the patchouli shoot culture on the TIS RITA bioreactor in different immersion time. In bioreactor of TIS RITA 1, patchouli shoots were submerged in medium for 5 minutes followed by 1 hour off media (5: 1), TIS RITA 2 (15: 1), and TIS RITA 3 (30: 1). The result showed that shoots biomass increase three times, but not different between treatments and there was a morphological difference in the resulting plantlets. TIS RITA 1 bioreactor produced highest root dry weight.TIS RITA 2 produced highest leaf dry weight, and TIS RITA 3 produced highest stem dry weight. Patchouli oil was extracted using steam distillation from stems and leaves. Yield of essential oil of TIS RITA 1, TIS RITA 2, and TIS RITA 3 was 3.7%±0.34%; 3%±0.15%; 3.3%±0.04%. The composition of patchouli oil was heterogenous, but no patchouli alcohol was found in all patchouli oil samples. <br />
