Description
Summary:

Patchouli is a herbaceous plant used as raw material of patchouli oil. One of the problems in patchouli cultivation is the heteroginity of plant quality, causing inconsistency of patchouli oil quality. This might be caused by low quality seeds and simple planting systems. High quality seed propagation can be prepared by in vitro shoots micropropagation. Patchouli quality can also be improved by regulating biotic and abiotic conditions around the plant, one of the examples is the presence of nutrients. Potassium (K) is known as one of the elements that play an important role in the physiological processes of plants and also one of the macronutrients needed by plants. This study was conducted by cultivating the patchouli shoot culture in thin layer (TLC)by using batch and adding potassium in the form of KNO3 with five different concentrations of 5; 7.5; 10; 12.5; And 15 mM on ½ MS0 medium. In sequence, the resulting dry weight of explants is 0.132; 0.136; 0.14; 0.122; And 0.13 gram with a growth rate of 0.0282; 0.0292; 0, 303; 0.0253; And 0.0276 days-1. Statical analysis of biomass was done by One Way ANOVA (α = 0,05) and post-hoc Tukey HSD test, the analysis resulted the significant difference between treatments, and the best result was achived by adding 10 mM Potassium. The explants was extracted by steam distillation and yielded crude extracts with different chemical composition between treatments, however there was no significant difference in oil content.
