Silybin is known to have biological activity as anti-cancer, antiinflammatory, hepatoprotective, antilipoperoksidative and chelating agents. Based on the retrosynthetic analysis, silybin can be synthesized from the eugenol, an essential oil from cloves. The first stage of the total synthesis of silybin is the synthesis of hydroxychavicol that can be obtained by demethylation eugenol. In this research demethylation of eugenol has been done with zeolite (ZSM-5) as catalyst, 48% solution of hydrogen bromide and solids LiCl. The reaction were monitored by TLC and each reaction was optimized. Crude product was purified using gravity column chromatography with the eluent n-hexane, n-hexane: ethyl acetate (9: 1), and n-hexane: ethyl acetate (8: 2). The product was then characterized by spectroscopic methods namely FTIR, NMR, UV and MS. Hydroxychavicol successfully synthesized and purified with a yield of 18,1% Rf value 0,37 (n-hexane: ethyl acetate, 8: 2). text
