The purpose of this research is to develop liver fibrosis model and to investigate the hepatoprotective activity of saponin fraction of Luffa acutangula (L.) Roxb seeds. Fibrosis model was developed in male Wistar rats - induced with CCl4 (i.p.) two times a week for eigth weeks. Liver tissue damage at 6th, 7th and 8th week was determined by organ indexes and HE, SR, and DAB stainings. Result showed that rats were given with saponin fraction at a dose of 10 mg/kg bw showed an increased in the spleen index compared to control (0,17 + 0,03 vs 0,20 + 0,03). In addition, administration of saponin fraction at doses of 10 mg/kg bw twice a week, 20 mg/kg bw twice a week, and 20 mg/kg bw once daily were given significant recovery on morphological liver observed macroscopically as well as microscopically. Moreover, the collagen expression and inflammatory cells observed in treated groups with Sirius Red and DAB stainings at 6th, 7th and 8th weeks were decreased compared to control group. Based on this data, we conclude that saponin fraction at a dose of 20 mg/kg bw once daily is the most effective as a hepatoprotector. text
