However, its production is limited due to a long time process, occurred very randomly, and decreasing number of gaharu producing trees such as Aquilaria malaccensis in nature. Therefore, this research was established in in vitro condition to evaluate the production of secondary metabolites in Aquilaria malaccensis shoot culture upon elicitation with methyl jasmonate (MeJA) and crude extract of Fusarium solani strain Gorontalo (GSL1) or Jambi (GSL2). Elicitation was performed by adding 100 µM MeJA or 8 mg/L fungal extract to A. malaccensis shoot culture grown in ½ MS liquid medium for 15 days. Secondary metabolite profile was analyzed by Gas Chromatography – Mass Spectrometry (GC-MS). In addition, A. malaccensis response to elicitors was observed through glucanase (AmGLU) gene expression. GC-MS result showed that elicitation induces the production of secondary metabolites which were not detected in non-elicited control groups. This included the production of benzene, 1,3-bis(1,1dimethylethyl); styrene; hexadecanoic acid, methyl ester and D-limonene with highest relative content found in GSL2 (0,3%; 0,99%; 0,29% and 0,49%, respectively). Nevertheless, several compounds were also detected in both treated and non-treated groups with highest relative content also found in GSL2, such as octadecane 0,54%; dibutyl phtalate 3,48%; and phenol, 2,4-bis(1,1-dimethylethyl)- 0,78%. Additional analysis with AmGLU gene showed that expression level of AmGLU in treatment groups increased 16 hours after elicititation, while control group had relatively stable gene expression. The highest uplift observed in GSL2 that was almost doubled from initial expression level. This result showed that AmGLU mediates early response to elicitor by increasing gene expression following by accumulation of secondary metabolites. text
