Summary:The study was conducted to examine the effect of different concentration of nitrogen on growth kinetics, astaxanthin content, and astaxanthin productivity in H. pluvialis culture using Rudic medium in photobioreactor. The cultivation has two stages, green and red stage. At green stage, cultures were cultivated for 8 days in 1 L bottle, using Rudic medium, pH 6-9, 3000 lux (bright: dark, 24: 0) florescent light, at room temperature of 20oC, and aeration rate 1,5 L/min. Treatment was performed by using different nitrogen concentration in the red stage phase, with NaNO3 concentration of 75 mg / L, 150 mg / L, 225 mg / L, 300 mg / L (control), 375 mg / L, and without NaNO3. The astaxanthin was analyzed by HPLC using column C18, mobile phase methanol:H2O (95:5 v/v), flow rate 1.25 ml/minute, and UV detector λ= 474 nm. The data obtained then tested with one-way ANOVA statistics. The result of this study indicated that nitrogen has no significant effect on growth kinetics, astaxanthin content, and astaxanthin productivity. The highest astaxanthin content (0.16 mg/g) was obtained in NaNO3 with concentration of 75 mg/L. The highest astaxanthin productivity of 9.7 mg/L.year was obtained in NaNO3 with concentration of 75 mg/L.
