them by inhibiting enzymes involved in carbohydrate hydrolysis process such as <br />
α-amylase which can delay glucose absorption. The population in Indonesia used <br />
<br />
plants as traditional drug for treatment diabetes, the example are Strobilanthes <br />
<br />
crispa L. Blume leaf, Spondias dulcis Soland ex. Park leaf, Foeniculum vulgare <br />
<br />
Mill. seed, Alstonia scolaris (L.) R.Br. stem bark, Blumea balsamifera (L.) DC. <br />
<br />
leaf, Dioscorea hispida Dennst. leaf, Ocimum sanctum L. leaf, Phaseolus vulgaris <br />
<br />
L. seed, Ceiba pentandra (L.) Gaertn stem bark, Morus alba L. leaf, Syzygium <br />
<br />
cumini Merr. seed and stem bark, Phyllanthus acidus (L.) Skeels fruit, <br />
<br />
Phyllanthus emblica L. leaf, Phyllanthus niruri L. herb, Plantago major L. leaf, <br />
<br />
Gardenia jasminoides J. Ellis leaf, and Guazuma ulmifolia Lamk. var. tomentosa <br />
<br />
K. Schum leaf. The purpose of this research was to verify the plants that have <br />
<br />
used traditionally as diabetes drugs with an in vitro approach for α-amylase <br />
<br />
enzyme inhibitor. Extraction was done by reflux method using 96% ethanol as <br />
<br />
solvent. The extracts obtained were freeze dried and tested their activity for α- <br />
<br />
amylase inhibition at a concentration of 500 μg/mL. Ceiba pentandra (L.) Gaertn <br />
<br />
stem bark extract had the highest presentage of inhibition compared to other test <br />
<br />
samples with a value of 97,64±0,52%. IC50 values of extract of P. emblica L. leaf, <br />
<br />
S.cumini Merr. seed, S. cumini Merr. stem bark, C. pentandra (L.) Gaertn stem <br />
<br />
bark were 35,56±8,09 μg/mL; 25,63±1,91 μg/mL; 21,84±0,24 μg/mL; 12,62±1,81 <br />
μg/mL; and 3,01±0,77 μg/mL respectively. Then C. pentandra (L.) Gaertn <br />
<br />
became the selected plant for futher testing. The C. pentandra (L.) Gaertn stem <br />
<br />
bark ethanol extract is fractionated by liquid-liquid extraction method using nhexane <br />
<br />
and ethyl acetate. IC50 values for the n-hexane fraction, ethyl acetate <br />
<br />
fraction, and water fraction of C. pentandra (L.) Gaertn. stem bark were <br />
<br />
491.41±29.80 μg/mL; 322.61±9.29 μg/mL; and 26.49±1.07 μg/mL respectively. <br />
<br />
The verification of 18 plants that traditional used for diabetes treatment was done, <br />
<br />
C. pentandra (L.) Gaertn. stem bark extract had the best activity compared with <br />
<br />
its fractions. Simplicia and extract of C. pentandra (L.) Gaertn. stem bark contain <br />
<br />
of alkaloids, flavonoids, condensed tannin, saponin, quinones and <br />
<br />
steroids/triterpenoids. |
