Summary:Stevia (Stevia rebaudiana Bertoni) has a great potential to be used as natural sweetener because it contains steviol glycosides that approximately 250-450 times sweeter than sucrose but low in calories. Micropropagation of stevia plants in vitro has been developed because it is considered more effective than ex vitro. One of the most widely used systems for plant micropropagation in vitro is the bubble column bioreactor. Large scale research on stevia plant has been done, but there is no large scale in vitro production system using bubble column bioreactor using stevia plant originating from Indonesia. The aim of this research was to determine the optimum air flow rate for the micropropagation of stevia shoot culture in bubble column bioreactor. Shoot culture was initiated on MS medium supplemented with kinetin 1 ppm and cultivated in a bubble column bioreactor for 14 days. The parameters measured were growth, consumption rate of sucrose and ion, and rebaudioside-A content with HPLC. The results showed that culture with 0.05 vvm air flow rate resulted in higher shoots growth with productivity 0.32 ± 0.119 g/L medium.day, compared with culture with flow rate 0.1 vvm (productivity 0.21 ± 0.024 g/L medium.day). Culture with 0.1 vvm flow rate resulted in a higher rebaudioside-A content of 97.89 mg/L, compared to the 0.05 vvm flow rate (69.66 mg/L). In this study, the kLa value was 3.567 x 10-12 on the culture of bubble column with 0,05 vvm and 1.092 x 10-11 in the culture of bubble column with 0,1 vvm, which showed a mismatch between increasing kLa value with the expected biomass increase which might be due to oxidative stress and shear stress in bubble column bioreactor with 0.1 vvm flow rate.
