This study aims to enhance lutein and zeaxanthin content and productivity using NPK fertilizer. The study was conducted by substitute the macronutrient compounds of N, P, and K in Bristol medium using NPK fertilizer.Variations of NPK fertilizer concentration used were 0,125 g/L, 0,250 g/L, 0,375 g/L, and 0,500 g/L. Cultivation was carried out for 12 days in a photobioreactor with photoperiodization 12:12, flow rate at 1.4 vvm, light intensity of 6.000 lux, pH maintained in 7–7,5, temperature 26-30˚C. Biomass of Chlorella pyrenoidosa was harvested and extracted using diethyl ether. The extract was analyzed using a spectrophotometer at λ 445 nm for lutein quantification and λ 451 nm for zeaxanthin quantification. The results of this study show that the substitution of N, P, K in Bristol medium using NPK fertilizer at concentration 0,500 g/L increasing lutein and zeaxanthin content in Chlorella pyrenoidosa. Addition of NPK produce the highest lutein content of 5,077 mg/g (140% higher than lutein in Bristol medium) and the highest zeaxanthin content of 4,133 mg/g (137% higher than zeaxanthin in Bristol medium). The maximum lutein productivity was 169 mg/L/year and maximum zeaxanthin productivity was 138 mg/L/year. text |
